ABOUT: The purpose of this study is to investigate whether growth hormone releasing hormone (GHRH), also called tesamorelin, may help reduce liver fat and improve associated cardiovascular risk in overweight men and women with increased liver fat. Many overweight and obese individuals have reduced growth hormone levels. We think this may contribute to increased liver fat, and we are researching whether augmenting growth hormone (GH) secretion by giving GHRH will help to decrease liver fat. GHRH is not approved by the Food and Drug Administration for this purpose, and this study is investigational. This study consists of up to 16 visits, 1 screening visit to determine eligibility and between 12-15 additional visits over the next 18 months for those who qualify and choose to participate. There are no overnight visits in this study. Study visits include evaluation of health history, liver biopsy, blood draw, full body DEXA scan, coronary CT angiography, and MRI scan of abdominal fat and liver fat. Participants will be compensated for their time and effort with up to $1290. To qualify for this study you must be between 18-65 years of age, and you must have increased weight (overweight or obesity). You may not be currently taking any diabetes medications or weight loss medications. Women who are pregnant or breastfeeding may not participate. Seeking Overweight Men and Women Ages 18-65 years old for a Research Study You may be eligible for a research study taking place at Massachusetts General Hospital. • Ages 18-65 years old. • Subjects will be randomly assigned to take a medication called growth hormone releasing hormone (Tesamorelin) or placebo for 12 months, and all subjects will receive growth hormone releasing hormone for 6 additional months. • Study involves up to 16 visits to Massachusetts General Hospital. There are no overnight visits. • Reimbursement for transportation provided.
